![]() Count the number of Tetrahymena in the 5μl and record.Using the 2-20μl micropipettor, transfer 5μl to a concavity slide and view under the compound microscope at 4x. Observe each well under the dissecting microscope and chose based on number of cells in the dilution which one to use.Take 100μl from dilution -3, mix and add it to the last well.Take 100μl from dilution -2, mix and add it to the third well.Take 100μl from dilution -1, mix and add it to the second well.Add 100μl from the stock to the first well and mix by pipetting up and down.Using the 200-2000μl micropipettor, fill 4 wells with 900μl of the culture media.It will be used in later labs and is a very important skill. Serial Dilutions are taking a sample and lowering the concentration in particular steps. The purpose of this experiment is to develop the skill of diluting and to count the number of Tetrahymena in a certain sample and calculating how many cells/milliliters there are. They are being dried and the ciliates will go into “hibernation” until rehydrated. In the future we will be using the collected soil to examine soil ciliates. The petri dish was stored under a fume hood in the back of the lab to dry and use later and the leftover soil was placed in a bucket with other bags. Place ziplock bag with leftover soil in bucket with others.Cover the bottom of the Petri dish with collected soil and place the dish under the fume hood.Write collection number (EFD32F18) on Petri dish top and bottom.Go to the base area around a tree in the assigned area and using a spoon, fill approximately half of a ziplock bag with soil from 3-4cm deep.Look in the soil collection powerpoint to figure out what collection area is specified for each section.Also the soil collected will be used in a later lab towards the end of the semester. The purpose of soil collection is to develop and understanding of where exactly soil ciliates tend to live in our environment and to learn how to collect them. ![]()
